Expert Writing (1997): Studying the Secondary Structure of Ribosomal RNA Using Atomic Force Microscopy
Abstract: Ribosomal RNA (rRNA) was isolated using a repeated freeze-thaw method, which effectively disrupted ribosomes at low temperatures. The rRNA remained intact and could be well dispersed on mica surfaces without the need for extraction, denaturation, or staining. This sample was then analyzed using atomic force microscopy (AFM). The imaging revealed that rRNA molecules exhibit a multi-branched rod-like structure with high regularity. Based on their size and morphology, the rRNA molecules were classified into three distinct types, corresponding to 28S, 18S, and 5S rRNA, as predicted by computational models. All of them share similar secondary structures. The structural data obtained from 28S, 18S, 5.8S, and 5S rRNAs supports the thermodynamically stable secondary structures previously proposed. This study provides direct visual evidence of rRNA architecture at the single-molecule level, enhancing our understanding of ribosome structure and function.
