It is commonly used in clinical diagnostics to assess neurological damage or injury, particularly in cases of brain trauma, stroke, or neurodegenerative diseases. Experimental Principle: The assay is based on the sandwich enzyme immunoassay technique. A microplate is pre-coated with a specific monoclonal antibody against human S-100B. After incubation with the sample, the target S-100B protein binds to the immobilized antibody. A horseradish peroxidase (HRP)-labeled secondary antibody is then added, forming a complex of antibody-antigen-enzyme-labeled antibody. Following thorough washing to remove unbound components, the substrate TMB (3,3',5,5'-tetramethylbenzidine) is introduced. Under the catalytic action of HRP, TMB turns blue, and the reaction is stopped by adding an acidic solution, which changes the color to yellow. The intensity of the yellow color is directly proportional to the concentration of S-100B in the sample. The optical density (OD) is measured at 450 nm using a microplate reader, and the concentration of S-100B is determined by comparing the sample OD value to a standard curve generated from known concentrations. This method ensures high specificity, sensitivity, and reproducibility, making it ideal for routine laboratory testing.
