Experimental analysis of zebrafish triiodothyronine (T3)

The Zebrafish Triiodothyronine (T3) Experimental Analysis Kit is designed to quantify the levels of triiodothyronine (T3) in zebrafish serum, plasma, and other biological fluid samples. This assay follows a double-antibody sandwich immunoassay principle. The process begins with coating a microplate with purified zebrafish T3-specific antibodies, creating a solid-phase antibody platform. After incubation, the sample containing T3 antigen is added, allowing it to bind to the immobilized antibodies. Subsequently, an HRP-conjugated anti-T3 antibody is introduced, forming a complex of antibody-antigen-enzyme-labeled antibody. Following a thorough wash to remove unbound components, the substrate TMB is added. Under the catalytic action of HRP, TMB turns blue and then shifts to yellow when an acidic stop solution is applied. The intensity of the color change is directly proportional to the concentration of T3 in the sample. A microplate reader measures the optical density (OD) at 450 nm, and the T3 concentration is determined by comparing the OD values against a pre-established standard curve. This method ensures high specificity and sensitivity for T3 detection in zebrafish samples, making it a reliable tool for endocrinological research and toxicological studies.
