**Principle of the Assay**
The assay is based on the double-antibody sandwich method. A microtiter plate is pre-coated with a specific antibody against ADM. After incubation with the sample, the ADM present in the sample binds to the immobilized antibody. An HRP-labeled secondary antibody then recognizes the captured ADM, forming a complex. Following washing steps, the TMB substrate is added, and the reaction is stopped with sulfuric acid. **Sample Preparation**

- **Urine**: Centrifuge at 2000–3000 rpm for 20 minutes.

- **Tissue samples**: Homogenize in PBS, centrifuge, and collect the supernatant.
**Assay Procedure**
1. Prepare standard dilutions and add to designated wells.
2. Add sample diluent and test samples to the appropriate wells.
3. Seal the plate and incubate at 37°C for 30 minutes.
4. Wash the plate five times with diluted washing buffer.
5. Add HRP-conjugated reagent to each well (except blank).
6. Incubate again for 30 minutes.
7. Add TMB substrate and incubate for 15 minutes in the dark.
8. Stop the reaction with stop solution.
